Exendin‑4 is a 39‑amino‑acid peptide naturally derived from the saliva of the Gila monster (Heloderma suspectum). It is a long‑acting glucagon‑like peptide‑1 (GLP‑1) receptor agonist with high sequence homology to mammalian GLP‑1. Unlike endogenous GLP‑1, Exendin‑4 is resistant to degradation by dipeptidyl peptidase‑4 (DPP‑4), resulting in prolonged biological half‑life. It stimulates insulin secretion, suppresses glucagon release, slows gastric emptying and reduces food intake, making it widely used in research on diabetes mellitus, obesity and metabolic disorders. Recombinant Exendin‑4 is non‑glycosylated, commonly produced in Escherichia coli, and serves as an essential tool protein for GLP‑1 pathway studies and drug screening.
