Enhanced Green Fluorescent Protein (eGFP) is an engineered variant of wild‑type GFP originally derived from Aequorea victoria. Compared with native GFP, eGFP contains two key amino acid mutations (F64L and S65T), which significantly improve folding efficiency, fluorescence intensity and photostability, and accelerate chromophore maturation at 37 °C. It exhibits strong green fluorescence with excitation at ~488 nm and emission at ~509 nm. As a widely used reporter and marker protein, recombinant eGFP is commonly expressed in E. coli with high purity. It can be applied in protein localization, molecular interaction studies, fluorescence calibration, positive control for expression system validation, and quality control of fluorescence‑related detection platforms.
